Uncertain significance for Familial hypobetalipoproteinemia 1; Hypercholesterolemia, autosomal dominant, type B — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000384.3(APOB):c.7209G>T (p.Lys2403Asn), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the APOB gene (transcript NM_000384.3) at coding-DNA position 7209, where G is replaced by T; at the protein level this means replaces lysine at residue 2403 with asparagine — a missense variant. Submitter rationale: This sequence change replaces lysine with asparagine at codon 2403 of the APOB protein (p.Lys2403Asn). The lysine residue is weakly conserved and there is a moderate physicochemical difference between lysine and asparagine. This variant is present in population databases (rs777497625, ExAC 0.002%). This variant has not been reported in the literature in individuals affected with APOB-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
