Uncertain significance for Cardiomyopathy, familial restrictive, 3; Hypertrophic cardiomyopathy 2; Dilated cardiomyopathy 1D — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001276345.2(TNNT2):c.247A>G (p.Asn83Asp), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the TNNT2 gene (transcript NM_001276345.2) at coding-DNA position 247, where A is replaced by G; at the protein level this means replaces asparagine at residue 83 with aspartic acid — a missense variant. Submitter rationale: This sequence change replaces asparagine with aspartic acid at codon 73 of the TNNT2 protein (p.Asn73Asp). The asparagine residue is highly conserved and there is a small physicochemical difference between asparagine and aspartic acid. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a TNNT2-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, this variant is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance.
