Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.3530A>C (p.Gln1177Pro), citing Ambry Variant Classification Scheme 2023: The p.Q1177P variant (also known as c.3530A>C), located in coding exon 23 of the ALK gene, results from an A to C substitution at nucleotide position 3530. The glutamine at codon 1177 is replaced by proline, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
