Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000222.3(KIT):c.833G>C (p.Ser278Thr), citing Ambry Variant Classification Scheme 2023: The p.S278T variant (also known as c.833G>C), located in coding exon 5 of the KIT gene, results from a G to C substitution at nucleotide position 833. The serine at codon 278 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
