NM_000222.3(KIT):c.440C>T (p.Ser147Phe) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S147F variant (also known as c.440C>T), located in coding exon 3 of the KIT gene, results from a C to T substitution at nucleotide position 440. The serine at codon 147 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
