Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000222.3(KIT):c.1205C>T (p.Ala402Val), citing Ambry Variant Classification Scheme 2023: The p.A402V variant (also known as c.1205C>T), located in coding exon 7 of the KIT gene, results from a C to T substitution at nucleotide position 1205. The alanine at codon 402 is replaced by valine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
