Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.106A>T (p.Ser36Cys), citing Ambry Variant Classification Scheme 2023: The p.S36C variant (also known as c.106A>T), located in coding exon 1 of the KIF1B gene, results from an A to T substitution at nucleotide position 106. The amino acid change results in serine to cysteine at codon 36, an amino acid with dissimilar properties. However, this change occurs in the last base pair of coding exon 1, which makes it likely to have some effect on normal mRNA splicing. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
