Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000090.4(COL3A1):c.3098A>T (p.Asp1033Val), citing Ambry Variant Classification Scheme 2023: The p.D1033V variant (also known as c.3098A>T), located in coding exon 43 of the COL3A1 gene, results from an A to T substitution at nucleotide position 3098. The aspartic acid at codon 1033 is replaced by valine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
