Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.4028A>G (p.Tyr1343Cys), citing Ambry Variant Classification Scheme 2023: The p.Y1297C variant (also known as c.3890A>G), located in coding exon 35 of the KIF1B gene, results from an A to G substitution at nucleotide position 3890. The tyrosine at codon 1297 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
