Pathogenic for Ehlers-Danlos syndrome, type 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000090.4(COL3A1):c.81del (p.Val28fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change deletes 1 nucleotide from exon 2 of the COL3A1 mRNA (c.81delT), causing a frameshift at codon 28. This creates a premature translational stop signal (p.Val28Leufs*38) and is expected to result in an absent or disrupted protein product. While this particular variant has not been reported in the literature, loss-of-function variants in COL3A1 are known to be pathogenic (PMID: 24922459). For these reasons, this variant has been classified as Pathogenic.
