Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001378454.1(ALMS1):c.8180C>G (p.Ser2727Cys), citing Ambry Variant Classification Scheme 2023: The p.S2728C variant (also known as c.8183C>G), located in coding exon 10 of the ALMS1 gene, results from a C to G substitution at nucleotide position 8183. The serine at codon 2728 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
