Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001378454.1(ALMS1):c.7817C>A (p.Ser2606Tyr), citing Ambry Variant Classification Scheme 2023: The p.S2607Y variant (also known as c.7820C>A), located in coding exon 10 of the ALMS1 gene, results from a C to A substitution at nucleotide position 7820. The serine at codon 2607 is replaced by tyrosine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
