NM_001035.3(RYR2):c.12215C>T (p.Thr4072Met) was classified as Uncertain Significance for Catecholaminergic polymorphic ventricular tachycardia by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015. This variant lies in the RYR2 gene (transcript NM_001035.3) at coding-DNA position 12215, where C is replaced by T; at the protein level this means replaces threonine at residue 4072 with methionine — a missense variant. Submitter rationale: Variant of Uncertain Significance due to insufficient evidence: This missense variant is located in the cytoplasmic domain of the RYR2 protein. Computational prediction tools and conservation analyses are inconclusive regarding the impact of this variant on the protein function. Computational splicing tools suggest that this variant may not impact the RNA splicing. To our knowledge, functional assays have not been performed for this variant nor has this variant been reported in individuals affected with cardiovascular disorders in the literature. This variant has been identified in 9/275082 chromosomes (9/34300 Latino chromosomes) in the general population by the Genome Aggregation Database (gnomAD). Available evidence is insufficient to determine the pathogenicity of this variant conclusively.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
