NM_001378454.1(ALMS1):c.12458A>T (p.Lys4153Ile) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K4154I variant (also known as c.12461A>T), located in coding exon 22 of the ALMS1 gene, results from an A to T substitution at nucleotide position 12461. The lysine at codon 4154 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
