This variant lies in the CBL gene (transcript NM_005188.4) at coding-DNA position 2360, where G is replaced by A; at the protein level this means replaces arginine at residue 787 with histidine — a missense variant. Submitter rationale: Variant summary: CBL c.2360G>A (p.Arg787His) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 6e-05 in 251462 control chromosomes (gnomAD). To our knowledge, no occurrence of c.2360G>A in individuals affected with Noonan Syndrome-Like Disorder and no experimental evidence demonstrating its impact on protein function have been reported. Two ClinVar submitters (evaluation after 2014) cite this variant as likely benign. Based on the evidence outlined above, the variant was classified as uncertain significance.
