NM_006147.4(IRF6):c.314T>C (p.Met105Thr) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.314T>C (p.M105T) alteration is located in exon 4 (coding exon 2) of the IRF6 gene. This alteration results from a T to C substitution at nucleotide position 314, causing the methionine (M) at amino acid position 105 to be replaced by a threonine (T). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. This missense alteration is located in a region that has a low rate of benign missense variation (Lek, 2016; Firth, 2009). This alteration is predicted to be deleterious by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
