Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.181C>A (p.Pro61Thr), citing Ambry Variant Classification Scheme 2023: The p.P61T variant (also known as c.181C>A), located in coding exon 1 of the ALK gene, results from a C to A substitution at nucleotide position 181. The proline at codon 61 is replaced by threonine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
