Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001378454.1(ALMS1):c.8306C>T (p.Ser2769Phe), citing Ambry Variant Classification Scheme 2023: The p.S2770F variant (also known as c.8309C>T), located in coding exon 10 of the ALMS1 gene, results from a C to T substitution at nucleotide position 8309. The serine at codon 2770 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
