Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_024529.5(CDC73):c.1016C>G (p.Pro339Arg), citing Ambry Variant Classification Scheme 2023: The p.P339R variant (also known as c.1016C>G), located in coding exon 11 of the CDC73 gene, results from a C to G substitution at nucleotide position 1016. The proline at codon 339 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
