Pathogenic for Parathyroid carcinoma — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_024529.5(CDC73):c.188T>C (p.Leu63Pro), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine, which is neutral and non-polar, with proline, which is neutral and non-polar, at codon 63 of the CDC73 protein (p.Leu63Pro). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with clinical features of familial isolated hyperparathyroidism (PMID: 18755853, 32761341; Invitae). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 403886). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt CDC73 protein function with a positive predictive value of 80%. For these reasons, this variant has been classified as Pathogenic.
