Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001382430.1(AKT1):c.34C>A (p.Leu12Met), citing Ambry Variant Classification Scheme 2023: The p.L12M variant (also known as c.34C>A), located in coding exon 1 of the AKT1 gene, results from a C to A substitution at nucleotide position 34. The leucine at codon 12 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
