NM_003545.4(H4C5):c.107G>A (p.Arg36Gln) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.107G>A (p.R36Q) alteration is located in exon 1 (coding exon 1) of the HIST1H4E gene. This alteration results from a G to A substitution at nucleotide position 107, causing the arginine (R) at amino acid position 36 to be replaced by a glutamine (Q). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. The in silico prediction for this alteration is inconclusive. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
