NM_000834.5(GRIN2B):c.3319C>G (p.Leu1107Val) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.3319C>G (p.L1107V) alteration is located in exon 13 (coding exon 12) of the GRIN2B gene. This alteration results from a C to G substitution at nucleotide position 3319, causing the leucine (L) at amino acid position 1107 to be replaced by a valine (V). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. This missense alteration is located in a region that has a low rate of benign missense variation (Lek, 2016; Firth, 2009). The in silico prediction for this alteration is inconclusive. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
