Uncertain significance — the classification assigned by Ambry Genetics to NM_001130009.3(GEN1):c.44A>C (p.Gln15Pro), citing Ambry Variant Classification Scheme 2023: The p.Q15P variant (also known as c.44A>C), located in coding exon 1 of the GEN1 gene, results from an A to C substitution at nucleotide position 44. The glutamine at codon 15 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
