Uncertain significance — the classification assigned by Ambry Genetics to NM_001130009.3(GEN1):c.58C>G (p.Arg20Gly), citing Ambry Variant Classification Scheme 2023: The p.R20G variant (also known as c.58C>G), located in coding exon 1 of the GEN1 gene, results from a C to G substitution at nucleotide position 58. The arginine at codon 20 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
