Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001308093.3(GATA4):c.1051T>G (p.Ser351Ala), citing Ambry Variant Classification Scheme 2023: The p.S350A variant (also known as c.1048T>G), located in coding exon 5 of the GATA4 gene, results from a T to G substitution at nucleotide position 1048. The serine at codon 350 is replaced by alanine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
