NM_032638.5(GATA2):c.138C>A (p.Asp46Glu) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D46E variant (also known as c.138C>A), located in coding exon 1 of the GATA2 gene, results from a C to A substitution at nucleotide position 138. The aspartic acid at codon 46 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
