NM_001999.4(FBN2):c.8210T>C (p.Met2737Thr) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M2737T variant (also known as c.8210T>C), located in coding exon 64 of the FBN2 gene, results from a T to C substitution at nucleotide position 8210. The methionine at codon 2737 is replaced by threonine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
