This variant lies in the PALB2 gene (transcript NM_024675.4) at coding-DNA position 1114, duplicating one base; at the protein level this means shifts the reading frame starting at serine residue 372, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This duplication of one nucleotide in PALB2 is denoted c.1114dupA at the cDNA level and p.Ser372LysfsX2 (S372KfsX2) at the protein level. The normal sequence, with the base that is duplicated in brackets, is GGAA[dupA]GTGA. The duplication causes a frameshift which changes a Serine to a Lysine at codon 372, and creates a premature stop codon at position 2 of the new reading frame. Although this variant has not, to our knowledge, been reported in the literature, it is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. We consider this variant to be pathogenic.
