Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_020937.4(FANCM):c.344T>C (p.Leu115Pro), citing Ambry Variant Classification Scheme 2023: The p.L115P variant (also known as c.344T>C), located in coding exon 1 of the FANCM gene, results from a T to C substitution at nucleotide position 344. The leucine at codon 115 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
