Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_020207.7(ERCC6L2):c.1232T>G (p.Leu411Trp), citing Ambry Variant Classification Scheme 2023: The p.L411W variant (also known as c.1232T>G), located in coding exon 7 of the ERCC6L2 gene, results from a T to G substitution at nucleotide position 1232. The leucine at codon 411 is replaced by tryptophan, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
