Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_194248.3(OTOF):c.1621G>A (p.Gly541Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 541 of the OTOF protein (p.Gly541Ser). This variant is present in population databases (rs397515435, gnomAD 0.003%). This missense change has been observed in individual(s) with auditory neuropathy and/or sensorineural hearing loss (PMID: 2575033, 26778470, 34416374, 38224868, 38378725). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 40177). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant.
