Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001375380.1(EBF3):c.952A>G (p.Arg318Gly), citing Ambry Variant Classification Scheme 2023: The c.925A>G (p.R309G) alteration is located in exon 10 (coding exon 10) of the EBF3 gene. This alteration results from an A to G substitution at nucleotide position 925, causing the arginine (R) at amino acid position 309 to be replaced by a glycine (G). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. This missense alteration is located in a region that has a low rate of benign missense variation (Lek, 2016; Firth, 2009). The in silico prediction for this alteration is inconclusive. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
