Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_022552.5(DNMT3A):c.833G>A (p.Gly278Asp), citing Ambry Variant Classification Scheme 2023: The p.G278D variant (also known as c.833G>A), located in coding exon 6 of the DNMT3A gene, results from a G to A substitution at nucleotide position 833. The glycine at codon 278 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
