NM_012144.4(DNAI1):c.659A>G (p.Asn220Ser) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N220S variant (also known as c.659A>G), located in coding exon 8 of the DNAI1 gene, results from an A to G substitution at nucleotide position 659. The asparagine at codon 220 is replaced by serine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
