Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001369.3(DNAH5):c.5987G>A (p.Cys1996Tyr), citing Ambry Variant Classification Scheme 2023: The p.C1996Y variant (also known as c.5987G>A), located in coding exon 36 of the DNAH5 gene, results from a G to A substitution at nucleotide position 5987. The cysteine at codon 1996 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
