Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001369.3(DNAH5):c.3904C>G (p.Leu1302Val), citing Ambry Variant Classification Scheme 2023: The p.L1302V variant (also known as c.3904C>G), located in coding exon 25 of the DNAH5 gene, results from a C to G substitution at nucleotide position 3904. The leucine at codon 1302 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
