Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.1808C>A (p.Pro603His), citing Ambry Variant Classification Scheme 2023: The p.P603H variant (also known as c.1808C>A), located in coding exon 10 of the DICER1 gene, results from a C to A substitution at nucleotide position 1808. The proline at codon 603 is replaced by histidine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
