NM_177438.3(DICER1):c.3655A>C (p.Ile1219Leu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I1219L variant (also known as c.3655A>C), located in coding exon 20 of the DICER1 gene, results from an A to C substitution at nucleotide position 3655. The isoleucine at codon 1219 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
