Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_016222.4(DDX41):c.13G>A (p.Glu5Lys), citing Ambry Variant Classification Scheme 2023: The p.E5K variant (also known as c.13G>A), located in coding exon 1 of the DDX41 gene, results from a G to A substitution at nucleotide position 13. The glutamic acid at codon 5 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
