Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_016222.4(DDX41):c.1095C>A (p.Phe365Leu), citing Ambry Variant Classification Scheme 2023: The p.F365L variant (also known as c.1095C>A), located in coding exon 10 of the DDX41 gene, results from a C to A substitution at nucleotide position 1095. The phenylalanine at codon 365 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
