Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001903.5(CTNNA1):c.2534A>G (p.Lys845Arg), citing Ambry Variant Classification Scheme 2023: The p.K845R variant (also known as c.2534A>G), located in coding exon 17 of the CTNNA1 gene, results from an A to G substitution at nucleotide position 2534. The lysine at codon 845 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
