NM_001903.5(CTNNA1):c.1573A>G (p.Lys525Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K525E variant (also known as c.1573A>G), located in coding exon 11 of the CTNNA1 gene, results from an A to G substitution at nucleotide position 1573. The lysine at codon 525 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
