Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000090.4(COL3A1):c.3577C>G (p.Pro1193Ala), citing Ambry Variant Classification Scheme 2023: The p.P1193A variant (also known as c.3577C>G), located in coding exon 48 of the COL3A1 gene, results from a C to G substitution at nucleotide position 3577. The proline at codon 1193 is replaced by alanine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
