NM_000088.4(COL1A1):c.2923C>G (p.Leu975Val) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L975V variant (also known as c.2923C>G), located in coding exon 40 of the COL1A1 gene, results from a C to G substitution at nucleotide position 2923. The leucine at codon 975 is replaced by valine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
