Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.976C>A (p.Leu326Ile), citing Ambry Variant Classification Scheme 2023: The p.L326I variant (also known as c.976C>A), located in coding exon 8 of the CHEK2 gene, results from a C to A substitution at nucleotide position 976. The leucine at codon 326 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
