Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_014679.5(CEP57):c.385C>G (p.Leu129Val), citing Ambry Variant Classification Scheme 2023: The p.L129V variant (also known as c.385C>G), located in coding exon 4 of the CEP57 gene, results from a C to G substitution at nucleotide position 385. The leucine at codon 129 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
