Uncertain significance — the classification assigned by Ambry Genetics to NM_016507.4(CDK12):c.3804G>T (p.Glu1268Asp), citing Ambry Variant Classification Scheme 2023: The p.E1268D variant (also known as c.3804G>T), located in coding exon 14 of the CDK12 gene, results from a G to T substitution at nucleotide position 3804. The glutamic acid at codon 1268 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
