Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001211.6(BUB1B):c.1396G>C (p.Asp466His), citing Ambry Variant Classification Scheme 2023: The p.D466H variant (also known as c.1396G>C), located in coding exon 10 of the BUB1B gene, results from a G to C substitution at nucleotide position 1396. The aspartic acid at codon 466 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
