NM_000059.4(BRCA2):c.7183C>G (p.His2395Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H2395D variant (also known as c.7183C>G), located in coding exon 13 of the BRCA2 gene, results from a C to G substitution at nucleotide position 7183. The histidine at codon 2395 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
